Metabolic imprinting in beef calves supplemented with creep feeding on performance, reproductive efficiency and metabolome profile

This experiment evaluated the influence of creep feeding supplementation on productive and reproductive performance and on serum metabolome profile in Nelore (Bos indicus) heifers. Female calves were assigned to treatments: Creep (n = 190), with ad libitum access to a nutritional supplement from 70 to 220 days after birth, or Control (n = 140), without supplementation. After weaning (Day 220), both groups followed the same pasture and nutritional management. Body weight (BW) and backfat thickness (BFAT) were measured over time. Blood samples were collected at 220 and 360 days for LC–MS/MS targeted metabolomics. On day 408, during the synchronization timed artificial insemination (TAI) protocol, reproductive status (RS: diameter of uterine horn and largest follicle, and presence of CL) was assessed. Creep feeding increased BW and BFAT at weaning, but no differences in BW, BFAT, or RS after weaning were observed. Nonetheless, the pregnancy per AI (P/AI) for 1st service was 28.9% higher in the Creep group. On day 220, 11 significant metabolites influenced five metabolic pathways: Glucose-alanine cycle, alanine, glutathione, phenylalanine and tyrosine metabolism, and urea cycle. On day 360, 14 significant metabolites influenced eight metabolic pathways: Malate-aspartate shuttle, arginine and proline metabolism, urea cycle, aspartate, beta-alanine, glutamate metabolism, ammonia recycling and citric acid cycle. In conclusion, creep feeding supplementation improved calf performance and induced metabolic changes at weaning and 360 days of age. Although heifers had similar productive performance and reproductive status, when submitted to TAI, those supplemented with creep feeding had greater P/AI.

Effects of short and long – term nutrition and progesterone supplementation on the success of fixed – time artificial insemination in the ewe

The success of fixed - time artificial insemination (AI) in the ewe is variable due to poor synchrony of estrus. We examined the effects of long-term nutrition (LTN; low, medium, high - 6 months), short-term nutrition (STN; 1.0 M, 1.5 M – 14 days) and progesterone supplementation (P; single pessary, replacement on Day 9) on synchrony and reproductive outcomes. High LTN advanced (P < 0.05) estrus, increased (P = 0.06) pregnancy (range 71.1 – 81.1%) and improved (P < 0.01) litter size (range 1.30 – 1.50). STN increased (P < 0.05) pregnancy (79.0 versus 72.3%) but not litter size or timing of estrus. A LTN x STN interaction (P < 0.01) for time of estrus indicates that the effects of LTN were moderated by STN depending on the level of LTN. Pessary replacement delayed (P < 0.05) the onset of estrus, improved synchrony but did not affect pregnancy or litter size. High LTN increased (P < 0.05) the number of large (≥ 3.8 mm) and medium - size follicles (2.0 – 3.7 mm) but the diameter of large follicles tended to be reduced (P = 0.08) on Day 12. STN did not affect follicle number or size whilst P reduced (P < 0.05) the diameter of large follicles on Day 12 (4.83 versus 5.10 mm) and increased the number of medium – size follicles (3.56 versus 2.74 mm). In conclusion, both LTN and STN are major sources of variability in AI programs whilst pessary replacement has potential to reduce variability.

Blood Plasma Biochemical Changes and Conception Rate in Anoestrus Buffaloes Treated with Various Hormone Protocols

To study certain blood biochemical parameters (total cholesterol, total protein, calcium and inorganic phosphorus) at various periods of treatment. Total 40 anestrous buffalo heifers with history of anoestrus were selected from the semi-arid area of Banaskanth and randomly divided into four equal groups. (1)Group-I:Heatsynch protocol (n=10), (2)Group-II:Ovsynch protocol(n=10), (3)Group-III: Doublesynch protocol (n=10) and (4)Group-IV: Control(n=10) followed by fixed time artificial insemination in treated buffalo heifers and insemination at spontaneous estrus in buffalo heifers of group-IV. The overall conception rate following 3 consecution services was maximum in Group II (50%) followed by Group III (40%), Group I (30%) and Group IV (30%). The blood plasma was collected at the time of initiation of treatment (P1), at the time of inj. PGF2α (P2), at the time of 1st FTAI (P3) and day 21st post insemination (P4) for the estimation of biochemical parameters. The blood biochemical profiles include total protein (range: 4.54± 0.19 to 6.11± 0.52 g/dl) which in all group did not show any significant difference during the different period but significant (P&lt;0.05) difference between the groups was noticed. Similarly calcium and phosphorus found ranging between 5.73± 0.20 to 9.53± 0.95 mg/dl and 5.46± 0.22 to 6.95± 0.30 mg/dl, respectively in all four groups. The cholesterol estimated values (range: 34.12 ± 1.75 to 41.97± 2.43 mg/dl) were found not-significantly varying between the groups as well as among the different periods. Heatsynch, Ovsynch and Doublesynch protocol can be successfully used for the induction of estrus in true anoestrus buffalo heifers with Better conception rate would be achieved following Ovsynch protocol in buffalo heifers.

Effect of a Single Injection of GnRH Analog Alone and at the Time of AI on Reproductive Performance of Nili Ravi Buffaloes

Introduction: The gonadotropin-releasing hormone (GnRH) is a decapeptide hormone released from basal neurons of the hypothalamus. It stimulates the production of follicle-stimulating hormone (FSH) and luteinizing hormone (LH)  in the pituitary. The objective of the study was to evaluate the effect of a single injection of gonadotropin-releasing hormone (GnRH) analog at the time of artificial insemination (AI) on buffalo estrus, conception, pregnancy, and calving rate. Materials and methods: The anestrus nulliparous Nili Ravi buffalo heifers (n=80) were divided into 4 groups randomly. Group A (n=20) received only a single injection of GnRH analog at start of the experiment (Day 0), group B (n=20) received a single injection of GnRH analog at Day 0 and prostaglandine (PGF2α) at Day 7, group C (n=20) received progesterone sponge for 7 days, and group D (n=20) received progesterone sponge for 7 days and PGF2α at Day 9. In 2nd study, buffaloes (n=60) expressing first (n=20), second (n=20), and third (n=20) postpartum estrus were enrolled. Moreover, heifers (n=20), expressing estrus first time were also included. Animals (n=10) from each group, received a single injection of GnRH analog at the time of AI were named as treatment group and the control (n=10) did not receive any treatment. Pregnancy tests were performed with the help of ultrasound on days 35 and 65 post-AI. Results: The pregnancy and calving rates were lower in GnRH group compared to other treatments. Similarly, the administration of GnRH analog did not increase pregnancy rates in treatment groups. Conclusion: The single injection of GnRH analog alone at the time of AI did not improve the reproductive performance of buffaloes.       

82 Effects of a red-clover containing mineral on heifer behavior and fertility

Abstract Heat stress reduces reproductive efficiency in beef cattle. Recent evidence suggests that supplementation of red clover can alleviate the physiological impacts of fescue toxicosis which could decrease heat stress. Our objective was to determine the effect of supplementing heifers grazing endophyte-infected tall fescue (E+ TF) with a conventional mineral containing red-clover leaf. We hypothesized that heifers supplemented with the red-clover leaf mineral would have greater conception rates to AI (CRAI) in times of possible heat stress compared with heifers supplemented with conventional mineral. A grazing study was conducted from May – August in 2021, 2022, and 2023. Yearling beef heifers (n = 32, 36, 40 heifers yr-1, respectively) were blocked by body weight, hair coat score and color into 4 grazing blocks that were assigned to 1 of 2 treatments (2 blocks treatment-1): 1. conventional non-medicated loose mineral control (0%-RC), or 2. conventional loose mineral + 20% wt/wt red clover leaf (20%-RC). Mineral treatments were provided in ground, 3-compartment mineral feeders ad libitum. Each grazing block was assigned to four, 0.8- ha. pastures consisting of predominantly E+ TF and were rotationally managed over the course of the study. Each year estrus was synchronized suitable for fixed time AI in May and all heifers resynchronized in June. In addition, each heifer was fitted with an ear-tag sensor (CowManager SensOor, Agis Automatisering BV, Harmelen, the Netherlands) for continuous behavioral monitoring (including high activity and activity). The CRAI were analyzed with the GLIMMIX procedure of SAS using the binomial distribution link logit function (v.9.4., SAS Inst. Inc., Cary, NC). Total daily activity and high activity reported by CowManger during May and June were analyzed by the MIXED procedure with repeated measures of SAS. The CRAI were similar for 20%-RC (67.30 ± 9.80%) and 0%-RC (60.70 ± 9.80%) heifers following the first service of AI (P = 0.51) during a thermoneutral period in May. In contrast, CRAI was greater (P = 0.02) for 20%-RC heifers (48.67 ± 13.47%) than CM (15.10 ± 13.47%) in June when temperatures increased to potential heat stress levels. Overall, conception rate after two rounds of AI for 20%-RC (78.30 ± 9.30%) heifers tended to be greater (P = 0.08) than CM (62.20 ± 9.30 %). While high activity (associated with estrous behavior) reported by CowManager did not differ between treatments (20%-RC: 203.94; 0%-RC: 213.90; SEM: 8.65 min d-1; P = 0.25), normal daily activity of 20%-RC heifers (150.69 ± 10.40 min d-1) tended to be greater (P = 0.06) than CM (126.33 ± 8.65 min d-1). Results of this study suggest supplementing red-clover containing mineral may mitigate negative effects of consuming E+ TF including decreased conception rates during times of heat stress.

Effect of 200 μg of gonadorelin hydrochloride at the first GnRH of a CIDR Synch program on ovulation rate and pregnancies per AI in Holstein heifers

The initial ovulatory response during synchronization programs is often low in dairy heifers, largely due to follicular dynamics and hormonal dynamics. Specifically, the progesterone concentration (P4) at the time of the first GnRH treatment in a breeding program can influence the LH response, often resulting in a suboptimal ovulatory response. The objective of this study was to determine the effect of the highest label dose 200 μg (100 μg vs. 200 μg) of GnRH (50 μg gonadorelin hydrochloride per mL; Factrel®; Zoetis Inc. Madison, NJ) at the first GnRH of a 6-d CoSynch plus P4 device program on ovulatory response and pregnancy per AI (P/AI) in first service in Holstein heifers. A total of 1308 Holstein heifers were randomly allocated at the beginning of a 6-d CIDR-Synch program, Day 0, to receive either i.m. treatment of 100 μg (2CC, n = 655) or 200 μg (4CC, n = 653) of GnRH. Also, at Day 0, heifers received an intravaginal insert with 1.38 g of P4 (Eazi-Breed CIDR® Cattle Insert; Zoetis Inc., Madison, NJ). On Day 6, the insert was removed, and i.m. treatment of 25 mg of PGF2α (12.5 mg dinoprost tromethamine/mL; Lutalyse® HighCon Injection Zoetis) was administered. On Day 7, a second i.m. treatment of 25 mg of PGF2α was given, followed on Day 9 by concurrent i.m. treatment of 100 μg of GnRH and timed AI (TAI). A subset of 396 heifers had their ovaries scanned to evaluate ovulatory response, and blood samples were collected to measure the serum concentration of P4 at Day 0 and Day 6 of the study. The P4 concentrations at Day 0 were categorized as Low (≤3ng/mL) or High (>3ng/mL). The ovulatory response was greater for heifers receiving 4CC than 2CC at Day 0 (54.7% vs. 42.8%). The ovulatory response was greater for Low P4 than High P4 at Day 0 (54.3% vs. 37.8%). However, there was not an interaction between treatment and P4 concentrations (Low P4 2CC = 48.6% vs. High P4 2CC = 30.0%; Low P4 4CC = 60.0% vs. High P4 4CC = 45.5%). The ROC curve analysis indicates that P4 concentrations at Day 0 treatment could predict the ovulatory response, although the area under the curve was only 0.6. As expected, heifers that ovulated had increased P/AI (No = 55.6% vs. Yes = 67.7%); however, there was no effect of treatment on P/AI (2CC = 63.3% vs. 4CC = 59.6%), nor interactions between treatment and ovulation and treatment and P4 (HIGH vs LOW) for pregnancy outcomes. In summary, P4 concentration and increasing the dose of GnRH at Day 0 positively impacted ovulatory response in Holstein heifers. However, there was no interaction between treatment and P4 on ovulation and no subsequent impact of GnRH dose on P/AI.

The Antioxidant Salidroside Ameliorates the Quality of Postovulatory Aged Oocyte and Embryo Development in Mice.

Postovulatory aging is known to impair the oocyte quality and embryo development due to oxidative stress in many different animal models, which reduces the success rate or pregnancy rate in human assisted reproductive technology (ART) and livestock timed artificial insemination (TAI), respectively. Salidroside (SAL), a phenylpropanoid glycoside, has been shown to exert antioxidant and antitumor effects. This study aimed to investigate whether SAL supplementation could delay the postovulatory oocyte aging process by alleviating oxidative stress. Here, we show that SAL supplementation decreases the malformation rate and recovers mitochondrial dysfunction including mitochondrial distribution, mitochondrial membrane potential (ΔΨ) and ATP content in aged oocytes. In addition, SAL treatment alleviates postovulatory aging-caused oxidative stress such as higher reactive oxygen species (ROS) level, lower glutathione (GSH) content and a reduced expression of antioxidant-related genes. Moreover, the cytoplasmic calcium ([Ca2+]c) and mitochondrial calcium ([Ca2+]mt) of SAL-treated oocytes return to normal levels. Notably, SAL suppresses the aging-induced DNA damage, early apoptosis and improves spindle assembly in aged oocytes, ultimately elevating the embryo developmental rates and embryo quality. Finally, the RNA-seq and confirmatory experience showed that SAL promotes protective autophagy in aged oocytes by activating the MAPK pathway. Taken together, our research suggests that supplementing SAL is an effective and feasible method for preventing postovulatory aging and preserving the oocyte quality, which potentially contributes to improving the successful rate of ART or TAI.

Supplementation with long-acting injectable progesterone 3days after TAI impaired luteal function in buffaloes.

Three experiments were conducted to evaluate the effects of long-acting injectable progesterone (iP4) in buffalo cows. In Experiment 1, ovariectomized buffaloes received 300mg (iP300) or 600mg (iP600) of iP4, and serum P4 concentrations were evaluated. In experiment 2, three groups were compared: control or administration of 300mg of iP4 3 (iP4-D3) or 6days (iP4-D6) after timed artificial insemination (TAI). On day 16, reproductive tract was recovered for conceptus, endometrium, and corpus luteum (CL) analysis. In experiment 3, pregnancy per AI (P/TAI) and proportion of pregnancy losses were evaluated after administration of 300mg of iP4 3 (iP4-D3) or 6days (iP4-D6) after TAI in lactating buffaloes. In experiment 1, serum P4 concentrations remained over 1ng/mL for ~ 3days in both groups. The 300mg dose was used in subsequent experiments. In experiment 2, CL weight and endometrial glands density were decreased, and conceptus length was increased in iP4-D3 compared to control and to iP4-D6 (P < 0.05). Transcript abundance of Prostaglandin F Receptor (FP) and ISG15 in CL and of ISG15 and MX1 in endometrium was greater in iP4-D3 when compared to control and to iP4-D6 (P < 0.05). In experiment 3, there was no difference among experimental groups for P/TAI at D30 and pregnancy losses (P > 0.1); however, iP4-D3 presented a lower P/TAI at day 60 (41.7%) when compared to control (56.8%) and iP4-D6 (57.7%; P = 0.07). In conclusion, administration iP4 at 3days after TAI affects CL development and consequently decreases final pregnancy outcome in buffaloes.

Economic viability of using OvSynch and fixed timed artificial insemination protocol in breeding improvement of pastoral herds in the rangelands.

Though using Assisted Reproductive Technologies (ARTs) can improve oestrus detection, conception and pregnancy success, thus benefit breeding program implementation, empirical evidence of their economic viability is lacking to inform investment decisions in pastoral herds. This study assessed economic viability of using OvSynch and fixed Timed Artificial Insemination (TAI) protocol in Sahiwal upgrading breeding program under two hypothetical cases of best and worst in activity-based money allocations when pastoral herds deploy either optimal or low input husbandry practices. From herd owners' assessment of milk production, best-case scenarios attain on average 10 L/cow/day with optimal husbandry and 5 L/cow/day with low input husbandry. The worst-case scenarios attain 5 L/cow/day with optimal husbandry and 1 L/cow/day with low input husbandry. Benefit- Cost Analysis (BCA) estimated Net Present Value (NPV), Benefit-Cost ratio (BCR) and Internal Rate of Return (IRR) to establish economic viability of using OvSynch and TAI Protocol in pastoral breeding programs. Both best-case scenarios retuned positive NPVs (82,028 and 6,912), BCR values (1.68 and 1.08) and IRR (27.46% and 8.08%) while worst-case scenarios returned negative NPVs (-135,855 and -141,025), BCR values of below 1 (0.87 and 0.66) and IRR values below the minimum rate of returns. These economic parameters were sensitive to price changes in inputs and outputs, under both optimal and low input husbandry practices. Results indicate that using OvSynch and TAI Protocol is a profitable and economically viable investment under optimal husbandry practices but not under low input husbandry practices. By implications, use of OvSynch and TAI Protocol in Sahiwal upgrading breeding programs need be accompanied with improved husbandry practices and de-risking pastoral herd owners from price changes in input and output markets.

Effects of equine chorionic gonadotropin dosage and its splitting in different days on reproductive performance of Nellore cows synchronized for timed-artificial insemination

This study evaluated the effects of dose of equine chorionic gonadotropin (eCG) and its splitting in different days of the synchronization protocol on reproductive performance of primiparous and multiparous Nellore cows. In the present study, 2,536 Nellore cows (1,634 primiparous and 902 multiparous) were assigned to receive in a 2 × 2 factorial design 1) an intravaginal progesterone (P4) device and 2.0 mg of estradiol benzoate (EB) on day −11, 12.5 mg (i.m.) of dinoprost tromethamine (PGF), 300 IU (i.m.) of eCG, 0.6 mg (i.m.) of estradiol cypionate (ECP), and P4 device withdrawal on day −2, followed by TAI on day 0 (n = 632 cows, being 409 primiparous and 223 multiparous; 300–2), 2) 300 IU (i.m) of eCG administered on days −4 and −2 (150 IU of eCG/day; n = 637 cows, being 412 primiparous and 225 multiparous; 300-4-2), 3) 400 IU (i.m.) of eCG administered on day −2 (n = 633 cows, being 406 primiparous and 227 multiparous; 400–2), and 4) 400 IU (i.m) of eCG administered on days −4 and −2 (200 IU of eCG/day; n = 634 cows, being 407 primiparous and 227 multiparous; 400-4-2). Individual cow BCS was assessed on days −11, 0 (timed-AI), and 31 of the study. Body condition score of the animals was classified into LOW or HIGH using the threshold of 2.75 (≤2.75 = LOW; >2.75 = HIGH). For primiparous cows, an eCG splitting effect was observed on follicle size, as cows receiving eCG on days −4 and −2 of the synchronization protocol had a larger follicle than cows administered eCG only on day −2. For day 31 P/AI, primiparous cows receiving 400-4-2, regardless of BCS, had a greater P/AI than cows from other treatments. Administering 400-4-2 to LOW BCS cows also resulted in greater P/AI than all other treatments assigned to LOW BCS cows. For multiparous cows, no treatment effect was observed for follicle size, estrus expression, and day 31 P/AI (P ≥ 0.21). In summary, increasing the dose and splitting the dose of eCG positively impacted the pregnancy rates of primiparous cows under a BCS ≤2.75, but no effects were detected on multiparous cows.

Effect of Double-Ovsynch and Presynch-Ovsynch on postpartum ovarian cysts and inactive ovary in high-yielding dairy cows.

Optimizing the management of dairy cattle reproduction can reduce postpartum ovarian disease in high-yielding dairy cows and thus enhance ranch economic benefit. The hypothesis of this study was that the Double-Ovsynch (DO) protocol in high-producing dairy cows would result in a lower incidence of follicular cysts but a higher incidence of luteal cysts compared to those undergoing the Presynch-Ovsynch (PS) protocol. In this experiment, 384 cows (204 primiparous and 180 multiparous) were allocated to the DO group, which followed the protocol: GnRH-7d-PGF2α-3d-GnRH-7d-Ovsynch-56 h (GnRH-7d-PGF2α-56 h-GnRH-16hTAI), starting on 39 ± 3 days in milk (DIM). Additionally, 359 cows (176 primiparous and 183 multiparous) were assigned to the PS group, which followed the protocol: PGF2α-14d-PGF2α-12d-Ovsynch-56 h, starting on 31 ± 3 DIM. In DO, B-mode ultrasound examinations were conducted 1 day after the GnRH-7d-PGF2α-3d-GnRH protocol to diagnose the presence of ovarian diseases followed by reexamination after 7 days of suspected cases. In PS, B-mode ultrasound examinations were conducted 1 day after the PGF2α-14d-PGF2α protocol to diagnose the presence of ovarian diseases followed by reexamination after 7 days. For all cows confirmed to having ovarian diseases, a second B-mode ultrasound examination was conducted at the time of the second GnRH and timed artificial insemination (TAI). If the ovary showed a normal developing follicle in combination with normal ovulation, the ovarian disease was considered to be cured. The current study revealed no significant difference in the overall incidence and cure rate of postpartum ovarian diseases between DO and PS (incidence rate: 3.9% vs. 6.7%, cure rate: 50% vs. 41.7%, DO vs. PS). Also, there was no significant difference in the incidence and cure rate of luteal cysts between DO and PS (incidence rate: 2.9% vs. 2.2%, cure rate: 50.0% vs. 50.0%). The incidence of follicular cysts was significantly lower in the DO group than in the PS group (0.8% vs. 2.8%, DO vs. PS, p = 0.037), but there was no significant difference in the cure rates (66.7% vs. 50%). The occurrence of inactive ovary was lower in DO compared to PS (0.2% vs. 1.7%, p = 0.047). There was no significant difference in the pregnancy rate between the DO and PS groups (48.2% vs. 41.8%), although the DO group had a higher rate. What is different from our assumption is that PS did not effectively reduce the incidence of postpartum luteal cysts.

Equine chorionic gonadotropin treatment and timed artificial insemination for dairy cow production under heat stress.

This study investigated the effects of timed artificial insemination (TAI) and equine chorionic gonadotropin (eCG) administration on lactating dairy cows under heat-stress conditions (average temperature-humidity index: 80). Timed artificial insemination was performed on the cows with (n = 57) or without (control, n = 41) supplementation with 500 IU of eCG at the day of PGF2α treatment using the CIDR-Ovsynch protocol. GnRH was administered, and a progesterone device (CIDR) was inserted on Day -10 of the treatment protocol. The CIDR was removed on Day -3, and the cows were treated with PGF2α. Two days later, a 2nd GnRH injection was administered. Subsequently, AI was performed on Day 0 (16-20 h after the 2nd GnRH injection), and pregnancy was diagnosed on Days 32 and 60. Plasma progesterone (P4) concentrations were measured after AI. Results showed that the eCG group had a higher pregnancy per AI (P/AI) than the control group (43.9 vs. 12.2%, P = 0.002), which was also accompanied by elevated P4 levels. Four cows in the eCG group had multiple calves, representing 7.0 and 16.0% of the group and pregnant cows, respectively. In conclusion, 500 IU of eCG combined with CIDR-Ovsynch in lactating dairy cows under severe heat stress conditions successfully improved fertility. However, the protocol may have a slight risk of multiple births.

Impact of presynchronization and Ovsynch on early postpartum ovarian activity and fertility of dairy cows in Libya.

Reproductive efficiency affects dairy cow profitability. Ovarian function in postpartum (P.P.) has been better understood using ultrasound and hormonal assays. Optimizing ovulation synchronization and carefully timing artificial insemination (TAI) can greatly enhance reproductive rates in dairy cows. This experiment was designed to investigate the reproductive performance and ovarian activity in early postpartum lactating dairy cows using the Presynch-PGF2α, Ovsynch protocol, and TAI. Randomly the cows were assigned to a control group and a treatment group, based on the chronological order of their calving date. On day 14 P.P., both groups received two cloprostenol treatments, 14 days apart. Ultrasonographic inspections were conducted on day 14 to check ovarian activity and uterine contents. On day 11, after presynchronization, cows in the treatment group were given 100 µg IM. of cystorelin, followed by a luteolytic dose of 500 µg IM., cloprostenol on day 7, and a second dose of cystorelin on day 8 (36 hours later). After the second cystorelin injection by 16-20 hours, cows were inseminated, while the control group had all cows displaying spontaneous estrus between day 0 and day 28 were artificially inseminated. Ovarian activity began to improve at 82.61% on day 19 P.P., with complete recovery between days 24 and 27 P.P. The second cloprostenol injection approached, causing follicular size to reach 8.41 ± 1.04 mm. After the second injection, ovarian activity switched from follicular to luteal, with corpus luteum rates of 23.91% and 26.1%. The presynchronized PGF2α regimen significantly enhanced ovarian activity from days 19-35 P.P. Ovulation and pregnancy rates in the Ovsynch group were 54.2% and 41.7% at the first timed artificial insemination (TAI), compared to 54.5% and 31.8% in the control group. There was no significant impact between them; it was just high in the presynchronized Ovsynch group. However, the P.P. period was minimized to 47-49 days till the first AI reached a 41.7% pregnancy rate and 20.8% at the second AI, for an overall 62.5%. The current study concludes that presynchronization during preservice in clinically normal P.P. dairy cows reduces P.P. duration, increases ovarian activity performance, and reduces ovarian dysfunctions from day 19 to day 35 P.P., as well as improves the pregnancy rate.

Social effects on behaviorally-scored and pedometer-detected estrus in beef cattle.

The number of cows in estrus often influences estrus behavior; however, the effects of social order are not well documented. This study examined the effects of social order on the expression of behaviorally-scored and pedometer-detected estrus, combined with the effects of the number of cows in estrus. In a herd comprising 13 or 15 beef cattle, cows with orders 1st-7th were defined as dominant and the remaining cows as subordinate. Sole or simultaneous estrus was induced by prostaglandin F2α analog injection and/or intravaginal progesterone treatment. Ovulation timing was determined using ultrasonography at 6-hour intervals. Estrous signs and steps of the cows were recorded 49 h before ovulation using video monitoring and a pedometer, respectively. Among the 59 treated cows, 56 behaviorally-scored estruses (27 sole and 29 simultaneous) were detected. In the sole estrus, 61.5% of the dominant-rank cows had no zero-point period; however, 35.7% of the subordinate-rank cows had that period. The dominant-rank cows in estrus alone had a significantly shorter duration of scored estrus than those in simultaneous estrus (P < 0.05). Among the 50 pedometer-detected estruses (24 sole and 26 simultaneous), the subordinate-rank cows in sole estrus had a shorter interval from estrus onset to ovulation than the dominant-rank cows in simultaneous estrus (P < 0.05). The effects of social order varied in response to the number of cows in estrus, which might have influenced determining the optimal time for artificial insemination.

Analysis of cow sweat volatile chemical compounds in gas chromatography mass spectrometry (GC-MS) device and their importance in heat detection

Detection of estrus is very important for the profitability of dairy cattle. Because one of the main goals of cattle breeding is for each cow to give birth to a healthy calf a year. The correct determination of the artificial insemination time to be applied to the animals by the enterprise is related to the correct determination of the animal’s estrus period. Knowing the signs of estrus that are specific to the estrus period of animals is very important in early and accurate detection of estrus. There are many methods for detecting estrus. One of the alternative methods to the methods developed to detect the estrus period in animals is the detection of volatile chemical compounds (pheromones) secreted in animal sweat only during the estrus period. This study will focus on the analysis of volatile chemical compounds in animal sweat using Gas Chromatography Mass Spectrometry (GC-MS) device and their importance in detecting estrus.

Influence of Ovarian Follicle Sizes and Estrous Signs on Pregnancy Following Progesterone-Based Fixed Time Artificial Insemination in Water Buffaloes

The objectives of the present study were to elucidate the importance of follicle sizes and estrous signs during Controlled Internal Drug Release-Synch-human Chorionic Gonadotropin (CIDR-Synch-hCG) protocol for Fixed Time Artificial Insemination (FTAI) and to evaluate their association with pregnancy in water buffaloes. Data from riverine buffaloes (n = 207) under the CIDR-Synch-hCG protocol were analyzed. Buffaloes were administered with Gonadotropin-Releasing Hormone (GnRH) with insertion of CIDR on Day 0. Prostaglandin (PGF2α) was given on Day 7 with the removal of CIDR. hCG was given on Day 9, and AI was performed on Day 10. Follicle measurements by ultrasonography were done on Days 0, 7, and 10, and follicle sizes on those days were categorized into I, II, and III. Estrus signs were taken on the day of AI. The pregnancy diagnosis was done on Day 30-35 post-AI. The average size of follicles in Category III is significantly higher than those of Categories I and II, regardless of the Days of the protocol. Pregnancy is significantly (P&lt;0.001) associated with Pre-Ovulatory Follicle (POF) size and uterine tonicity on the Day of AI but not with follicle sizes on Days 0 and 7, nor with mucus discharge discharge (P&gt;0.05). The overall pregnancy rate is 44.44% while performing AI with POF size ≥12.0 mm increased the probability of pregnancy rate to 56.25%. In conclusion, the present study demonstrated a follicle size-based CIDR-Synch-hCG protocol providing new fertility indicators to improve FTAI efficiency in buffaloes with huge application in other livestock species.

Fertility of Postpartum Iraqi Cows Following Timed Artificial Insemination within Ovsynch or Presynch Protocols

This investigation was carried out on privately owned dairy farms situated in Al-Anbar Province, Iraq, extending from June 2022 to March 2023. The objective of our study was to compare the calving interval (CI), days open (DO), and conception rate at first service (CRFS) between timed artificial insemination (TAI) within ovulation synchronization protocols and AI at estrus detection. Total cows (n = 80) 50–60 days postpartum were randomly divided. (1) AIED group (Control, n = 40) untreated cows received AI at the first observed estrus according to the a.m. /p.m. rule. (2) OVS group (n = 20) was treated with a classical Ovsynch protocol. (3) The PG+G group (n = 20) underwent resynchronization with PGF2 and GnRH at the same time 7 days later, then underwent an Ovsynch protocol. Pregnancy was confirmed using trans-rectal ultrasonography on day’s 30-35 post-insemination. The results of this study show that mean days of CI and DO for the control group were significantly longer than those in both the OVS group and the PG+G group (P &lt; 0.01). Among all groups, the CRFS showed a significant difference (37.5%, P &lt; 0.05). Separately, the CRFS for the control group was similar to the OVS group (respectively, 27.5% vs. 30%, P &gt; 0.05). Whereas, the PG+G group had a 65% CRFS, which was significantly higher than the control (P &lt; 0.01) and OVS (P &lt; 0.05). In summary, ovulation synchronization protocols enable TAI earlier in the postpartum period, eliminating the need for estrus detection and resulting in shorter CI and DO. Additionally, the PG+G protocol increased the CRFS compared to both the OVS and the control groups.
